About ABT ® GRAM STAINING KIT
Intended use: ABT ® Gram Staining Kit is utilized for bacterial staining to differentiate between two bacterial groups, Gram-positive (+) and Gram-negative (-), based on the structural composition of the bacterial cell wall.
Staining capabilities:
- Gram-positive bacteria exhibit a violet-green coloration
- Gram-negative bacteria display a pink-red hue
Storage: 15 - 30°C, protect from direct sunlight
Expiration date: 12 months from manufacture
Components: The kit includes 4 separate chemical bottles:
- Crystal violet solution: Homogeneous liquid slightly viscous liquid, with a purple or violet-green color. Exhibits a distinctive odor.
- Lugol solution: Homogeneous, moderately viscous liquid, with a reddish-brown color. Exhibits a distinctive odor.
- Decolorizer solution: Homogeneous, transparent, colorless liquid, free from sediment. Exhibits a distinctive odor.
- Safranin O solution: Homogeneous, slightly viscous liquid, red in color. Exhibits a distinctive odor.
Packaging: DD-002 (4 bottles x 100 mL/box)
Procedure

- Step 1: Evenly distribute the disease specimen or bacteria onto a clean microscope slide.
- Step 2: Fix the disease specimen by gently passing it over the flame of an alcohol lamp and allow it to cool.
- Step 3: First, apply Crystal Violet solution and let it stand for approximately 30 seconds, then rinse with water.
- Step 4: Next, apply Lugol solution to fix the stain for approximately 30 seconds, then rinse with water.
- Step 5: Decolor using Decolorizer solution for about 30 seconds, then rinse with water.
- Step 6: Finally, apply Safranin O solution for about 30 seconds, then rinse with water.
- Step 7: Allow to air-dry.
- Step 8: Observe under a microscope.
Note:
- Use gloves, goggles, a mask, and a lab coat during the procedure.
- Store the product in a dry, cool place, away from direct light, excessive heat, and potential sources of ignition. Close the product tightly after use.
- Immediately clean any stains with water to prevent permanent marks on sink surfaces, laboratory bench chairs, or glassware.
- Carefully read the instructions for use, pay attention to the expiration date of the product before use. Do not use the product after the expiration date.
- Dispose of waste in accordance with legal regulations and national environmental protection guidelines.
