About ABT ® Cell Preservative Solution
Intended use:
ABT ® Cell Preservative Solution is a medium used for the collection and preservation of gynecological cells for cytological analysis in cervical cancer screening (Pap test). Additionally, samples preserved in this medium can also be used for DNA extraction for HPV-DNA testing.
Cells preserved in ABT® Cell Preservative Solution under the action of alcohol and buffer salts will help fix and maintain their shape. Additionally, this solution inhibits the proliferation of microorganisms that are harmful to the cells, preserving the cells at room temperature for a period of time (at least 1 month).
Specification:
- Component: Methanol ≥ 35%

- Appearance: Homogeneous, clear, colorless liquid, free from impurities and sediment. Has a characteristic product odor, with no unusual smell

- Cell preservation capability: Captures and stabilizes the shape of nuclei and cytoplasm of gynecological cells at 15 – 25°C for 01 month
- Shelf life: 12 months from the date of manufacture
- Storage: Temperature 15 – 25°C

Procedure
- Sample Collection and Preservation
Cervical smear samples will be collected according to the Ministry of Health's procedure (Appendix 1, Decision 3877/QD-BYT 2019):
- Step 1: Place the duckbill into the vagina and adjust the position to ensure a clear view of the cervix.
- Step 2: Insert the sampling brush into the cervix, press gently, and rotate the brush clockwise 5 times.
- Step 3: Remove the sampling brush from the vagina and place it into the ABT® Cell Preservative Solution vial, gently rotating it 5-10 times. The head of the sampling brush is retained by detaching it from the handle, and the handle is disposed of properly.
- Step 4: Close the bottle cap and record the patient's information on the label.
Store and transport the sample at room temperature (15-25°C) for up to 1 month for cytological examination (Pap test) in cervical cancer screening. Additionally, the sample can also be used for DNA extraction for HPV-DNA testing.
- 2. Sample Processing for HPV-DNA Testing
Extracting and collecting DNA from cervical smear samples preserved in ABT® Cell Preservative Solution for HPV DNA tests, following the procedure below:
- Step 1: Draw 1 mL of the smear sample treated with ABT® Cell Preservative Solution into a 1.5 mL tube. Centrifuge at 10,000 rpm for 3 minutes, collect the pellet, and remove the supernatant.
- Step 2: Use the pellet for DNA extraction according to the procedure of the TopPURE® Genomic DNA Extraction Kit (HI-112) for the manual method or the TopPURE® Maga Genomic DNA/RNA Extraction Kit (HI-612) for the automatic method.
- Step 3: The extracted DNA will be used for qualitative or HPV typing tests following the instructions of the corresponding kits.
* Note:
– Shake the ABT® Cell Preservative Solution well before opening the cap for use.
– The appropriate sample collection time is within 2 days before or after menstruation.
– Contraindicated in pregnant women.
– Self-prepared materials: sampling brush, vaginal speculum, and 1.5 mL tube.
