About ABT ® Density Solution
Intended use:
ABT ® Density Solution is a medium used to eliminate obfuscating factors such as mucus, blood cells, and inflammatory cells, and to isolate target cells for cervical cytology screening (Pap test) via centrifugation.
Specification:
- Composition: polysaccharide, sodium azide, and alcohol.
- Appearance: Homogeneous, clear, colorless liquid, free from impurities and sediment. Has a characteristic product odor, with no unusual smell
- solution pH 5.5 ± 0.2
- Processing capability: Processes and removes mucus, blood, and inflammatory cells in gynecological smear samples (GYN) and collects target epithelial cells.
- Shelf life: 12 months from the date of manufacture
- Storage: Temperature 15 – 25°C
Packaging: 500mL/bottle (DD-104).

Procedure
- Sample Preparation: Cervical smear samples are collected and preserved in ABT® Cell Preservative Solution at 15-25°C for up to 1 month.
1. Manual Processing Procedure:
– Manually smear cervical smear samples using the Falcon Centrifuge according to the following procedure:
- Step 1: Vortex the ABT® Cell Preservative Solution vial containing the cervical smear sample for about 15 seconds.
- Step 2: Prepare and record the patient's information on a plastic pipette and a 15 mL Falcon tube.
- Step 3: Add 7 mL of the thoroughly mixed sample solution from Step 1 into the 15 mL Falcon tube.
- Step 4: Use a plastic pipette to draw 3 mL of ABT® Density Solution, insert the pipette straight to the bottom, and slowly release to allow the ABT® Density Solution to sink to the bottom and push the mixed sample solution up so that the two solutions form two separate layers.
- Step 5: Centrifuge at 4000 rpm for 10 minutes to separate the components in the smear sample.
- Step 6: Use a plastic pipette to remove approximately 6-7 mL of the supernatant. Centrifuge at 4000 rpm for 5 minutes to collect the biomass.
- Step 7: Use a plastic pipette to remove approximately 1-2 mL of the supernatant. Centrifuge at 4000 rpm for 5 minutes to completely pellet the biomass at the bottom of the falcon tube.
- Step 8: Completely remove the supernatant and collect the cell pellet to smear onto a glass slide.
2. Automatic Processing Procedure:
– Automatically smear cervical smear samples using the Cell Smear Centrifuge Machine (LTS-YJ1000) according to the following procedure:
- Step 1: Vortex the ABT® Cell Preservative Solution vial containing the cervical smear sample for about 15 seconds.
- Step 2: Prepare and record the patient's information on the frosted end of the glass slide of the centrifuge chamber.
- Step 3: Draw 2 mL of ABT® Density Solution and apply it to the surface of the settling chamber.
- Step 4: Draw 7 mL of the thoroughly mixed sample solution from Step 1 and slowly add it onto the surface of the ABT® Density Solution so that the two solutions form two separate layers.
- Step 5: Centrifuge at 1300 rpm for 2 minutes to smear the cells onto the glass slide.
- Step 6: Remove the supernatant and collect the glass slide.
