Introduction of ABT® PAPANICOLAOU STAINING KIT

Intended use: ABT ® Papanicolaou Staining Kit is engineered for the staining and morphological differentiation of gynecological cells in cervical smear preparations, widely utilized in screening for cervical cancer (which approximately 90% is attributed to HPV - Human Papillomavirus infection).

Expiration date: 12 months from manufacture

Components: The kit comprises three bottles of different staining solutions:

ABT® Nuclear Stain Solution: Liquid form, deep red in color, used for nuclear staining (Subsequent to staining, cell nuclei appear in violet).
ABT® OG Stain Solution: Liquid form, orange-golden in color, employed for cytoplasmic staining (After staining, cell cytoplasm appears in light orange).
ABT® EA Stain Solution: Liquid form, deep green with a pink hue, utilized for cytoplasmic staining (After staining, cell cytoplasm appears in shades of pink or light green).

Manual instruction

1. Prepare the staining reagents from the kit and other chemicals according to the following diagram:

Other equipment not provided: staining kit, glass slides.

Other chemicals not provided: Ethanol 96, washing liquid*.

Note:

Rinse water can be tap water with pH ≥ 8 or alkaline solutions such as Scott’s water, 0.05% Ammonia solution, or saturated lithium carbonate.
The staining reagents are supplied in Ready-to-use form; simply shake the staining reagent well and pour it into the container in the appropriate volume for use. Store in a tightly closed, opaque bottle after use.

2. Sample preparation:

Liquid-based Pap Test:

- The cervical fluid sample will be collected and processed following the procedure outlined in the TopPURE ® Pap Smear Collection Kit (HI-016).

- The liquid-based sample is transferred onto a microscope slide using an automated slide preparation machine and the corresponding filter cup.

- Traditional Pap Smear: The cervical fluid sample, after collection, will be directly smeared onto a microscope slide.

3. Papanicolaou Staining (PAP) Procedure:

- Step 1: Fix the sample with 96% Ethanol in box 1 for 15 minutes.
- Step 2: Dip the slide five times into the rinsing water in box 2 after fixation.
- Step 3: Stain the nuclei with ABT® Nuclear Stain Solution in box 3 for 1 minute.
- Step 4: Dip the slide five times into the rinsing water in box 2 after nuclear staining.
- Step 5: Successively dip the slide into 96% Ethanol in box 4 and 5. Repeat this process five times for each box.
- Step 6: Stain the cytoplasm with ABT® OG Stain Solution in box 6 for 1 minute.
- Step 7: Successively dip the slide into 96% Ethanol in box 7 and 8. Repeat this process five times for each box.
- Step 8: Stain the cytoplasm with ABT® EA Stain Solution in box 9 for 2 minutes.
- Step 9: Successively dip the slide into 96% Ethanol in box 10, 11, and 12. Repeat this process five times for each box.
- Step 10: Allow the slide to dry, apply mineral oil, and observe directly under the microscope.
