Add 400\u202f\u03bcl of <a href=\"https:\/\/abtvn.com\/en\/product\/phenol-saturated-ph-7-9\/\">Phenol saturated pH 7.9<\/a>, mix thoroughly, and centrifuge at 12,000 rpm for 1 minute.<\/li>\n<li>Collect the upper aqueous phase, add 800\u202f\u03bcl of <strong>Phenol: Chloroform: Isoamyl alcohol 25:24:1<\/strong> , mix well, and centrifuge at 12,000 rpm for 1 minute.<\/li>\n<li>Collect the upper aqueous phase, add 700\u202f\u03bcl of <a href=\"https:\/\/abtvn.com\/en\/product\/chloroform-isoamyl-alcohol-241\/\">Chloroform: Isoamyl alcohol 24:1<\/a>, mix thoroughly, and centrifuge at 12,000 rpm for 1 minute.<\/li>\n<li>Collect the upper aqueous phase, then add 1\/10 volume of 3\u202fM Sodium Acetate and 2 volumes of cold 96\u2013100% ethanol<\/li>\n<li>Mix gently until a precipitate forms. Centrifuge at 12,000 rpm, discard the supernatant, and wash the DNA pellet with 70% ethanol. Perform a minimum of 2 and up to 5 washes. After the final wash, remove residual ethanol and dry the pellet in a 50<span class=\"OYPEnA font-feature-liga-off font-feature-clig-off font-feature-calt-off text-decoration-none text-strikethrough-none\">\u00b0C<\/span> incubator for 15 minutes.<\/li>\n<li class=\"translation-block\">Add 500\u202f\u03bcl of 1X TE buffer. Incubate the tube in a water bath at 65\u201370\u00b0C for 30 minutes, or until the DNA is fully dissolved.<\/li>\n<\/ul>\n<h2><strong>Caution<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<ul>\n<li>Phenol is highly toxic upon contact with skin, eyes, ingestion, or inhalation. It may cause chemical burns.<\/li>\n<li>In case of skin contact, wash immediately with soap and plenty of clean water.
