M\u00e1y real-time PCR l\u1ea7n \u0111\u1ea7u ti\u00ean \u0111\u01b0\u1ee3c th\u01b0\u01a1ng m\u1ea1i h\u00f3a b\u1edfi Applied Biosystems v\u00e0o n\u0103m 1996, c\u00f4ng ngh\u1ec7 n\u00e0y nhanh ch\u00f3ng ph\u00e1t tri\u1ec3n th\u00e0nh m\u1ed9t th\u1ecb tr\u01b0\u1eddng c\u1ea1nh tranh, h\u00e0ng lo\u1ea1t c\u00e1c c\u00f4ng ty kinh doanh s\u1ea3n ph\u1ea9m sinh h\u1ecdc cho ra m\u1eaft c\u00e1c d\u00f2ng m\u00e1y real-time PCR.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">V\u1ec1 m\u1eb7t \u0111\u1ecbnh ngh\u0129a, k\u1ef9 thu\u1eadt real-time PCR cho ph\u00e9p bi\u1ebft \u0111\u01b0\u1ee3c s\u1ed1 l\u01b0\u1ee3ng b\u1ea3n sao DNA m\u1ee5c ti\u00eau t\u1ea1o ra ngay sau m\u1ed7i chu k\u1ef3 nhi\u1ec7t c\u1ee7a ph\u1ea3n \u1ee9ng b\u1eb1ng c\u00f4ng ngh\u1ec7 hu\u1ef3nh quang. Nguy\u00ean l\u00fd ho\u1ea1t \u0111\u1ed9ng c\u1ee7a k\u1ef9 thu\u1eadt real-time PCR<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">T\u01b0\u01a1ng t\u1ef1 nh\u01b0 PCR, k\u1ef9 thu\u1eadt real-time PCR c\u0169ng bao g\u1ed3m c\u00e1c th\u00e0nh ph\u1ea7n c\u01a1 b\u1ea3n nh\u01b0 dNTP, DNA Polymerase, DNA m\u1ea1ch khu\u00f4n, c\u1eb7p m\u1ed3i v\u00e0 dung d\u1ecbch \u0111\u1ec7m. \u0110i\u1ec3m kh\u00e1c bi\u1ec7t \u0111\u00f3 l\u00e0 real-time PCR s\u1eed d\u1ee5ng ch\u1ea5t ph\u00e1t hu\u1ef3nh quang \u0111\u1ec3 m\u00e1y c\u00f3 th\u1ec3 ph\u00e1t hi\u1ec7n v\u00e0 \u0111o \u0111\u01b0\u1ee3c c\u01b0\u1eddng \u0111\u1ed9 t\u00edn hi\u1ec7u t\u1eeb ch\u1ea5t n\u00e0y. Th\u1eddi gian bi\u1ebfn t\u00ednh c\u00f3 th\u1ec3 t\u0103ng l\u00ean n\u1ebfu th\u00e0nh ph\u1ea7n khu\u00f4n c\u00f3 nhi\u1ec1u GC.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">&#8211; Giai \u0111o\u1ea1n b\u1eaft c\u1eb7p: Nhi\u1ec7t \u0111\u1ed9 ph\u1ea3n \u1ee9ng gi\u1ea3m xu\u1ed1ng c\u00f2n 50-65<sup>0<\/sup>C trong 20-40 gi\u00e2y \u0111\u1ec3 probe v\u00e0 m\u1ed3i g\u1eafn l\u1ea7n l\u01b0\u1ee3t v\u00e0o s\u1ee3i DNA khu\u00f4n v\u00e0 b\u1eaft \u0111\u1ea7u qu\u00e1 tr\u00ecnh t\u1ed5ng h\u1ee3p m\u1ea1ch m\u1edbi nh\u1edd v\u00e0o s\u1ef1 ho\u1ea1t \u0111\u1ed9ng c\u1ee7a DNA polymerase.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">&#8211; Giai \u0111o\u1ea1n k\u00e9o d\u00e0i: Nhi\u1ec7t \u0111\u1ed9 ph\u1ea3n \u1ee9ng t\u0103ng l\u00ean 72<sup>0<\/sup>C, \u0111\u00e2y l\u00e0 nhi\u1ec7t \u0111\u1ed9 t\u1ed1i \u01b0u cho ho\u1ea1t \u0111\u1ed9ng t\u1ed5ng h\u1ee3p m\u1ea1ch m\u1edbi c\u1ee7a DNA polymerase. Giai \u0111o\u1ea1n n\u00e0y kh\u00f4ng b\u1eaft bu\u1ed9c \u1edf m\u1ed9t s\u1ed1 chu tr\u00ecnh qPCR, do k\u1ef9 thu\u1eadt n\u00e0y th\u01b0\u1eddng d\u00f9ng khu\u1ebfch \u0111\u1ea1i c\u00e1c \u0111o\u1ea1n c\u00f3 tr\u00ecnh t\u1ef1 ng\u1eafn h\u01a1n PCR, v\u00e0o kho\u1ea3ng &lt;200bp, do v\u1eady ch\u1ec9 c\u1ea7n hai b\u01b0\u1edbc bi\u1ebfn t\u00ednh v\u00e0 b\u1eaft c\u1eb7p l\u00e0 \u0111\u1ee7 \u0111\u1ec3 khu\u1ebfch \u0111\u1ea1i \u0111o\u1ea1n gen m\u1ee5c ti\u00eau trong ph\u1ea3n \u1ee9ng qPCR. Chu tr\u00ecnh ch\u1ec9 g\u1ed3m 2 b\u01b0\u1edbc gi\u00fap ti\u1ebft ki\u1ec7m th\u1eddi gian, nhanh ch\u00f3ng ph\u00e1t hi\u1ec7n v\u00e0 \u0111\u1ecbnh l\u01b0\u1ee3ng \u0111\u01b0\u1ee3c DNA.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">C\u00e1c giai \u0111o\u1ea1n n\u00e0y \u0111\u01b0\u1ee3c th\u1ef1c hi\u1ec7n th\u00f4ng qua s\u1ef1 lu\u00e2n chuy\u1ec3n nhi\u1ec7t \u0111\u1ed9 gi\u1eefa c\u00e1c chu k\u1ef3. Qua m\u1ed7i chu k\u1ef3, t\u00edn hi\u1ec7u hu\u1ef3nh quang s\u1ebd \u0111\u01b0\u1ee3c ghi nh\u1eadn c\u00f3 s\u1ef1 gia t\u0103ng t\u01b0\u01a1ng \u1ee9ng v\u1edbi l\u01b0\u1ee3ng b\u1ea3n sao DNA nh\u00e2n l\u00ean theo c\u1ea5p s\u1ed1 l\u0169y th\u1eeba.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>3. Th\u00e0nh ph\u1ea7n c\u1ee7a ph\u1ea3n \u1ee9ng real-time PCR<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Khi th\u1ef1c hi\u1ec7n real-time PCR, c\u00e1c th\u00e0nh ph\u1ea7n ph\u1ea3n \u1ee9ng s\u1ebd \u0111\u01b0\u1ee3c tr\u1ed9n v\u1edbi nhau trong tube v\u00e0 \u0111\u1eb7t trong m\u00e1y real-time PCR \u0111\u1ec3 th\u1ef1c hi\u1ec7n qu\u00e1 tr\u00ecnh nh\u00e2n b\u1ea3n DNA. Th\u00e0nh ph\u1ea7n ph\u1ea3n \u1ee9ng qPCR bao g\u1ed3m: m\u1eabu (khu\u00f4n DNA\/RNA), m\u1ed3i, ch\u1ea5t hu\u1ef3nh quang, \u0111\u1ec7m, DNA polymerase, nucleotide t\u1ef1 do (dNTP) v\u00e0 c\u00e1c ch\u1ea5t h\u1ed7 tr\u1ee3 (n\u1ebfu c\u1ea7n). Hi\u1ec7n nay c\u00f3 hai c\u00e1ch th\u1ee9c \u0111\u1ec3 chu\u1ea9n b\u1ecb cho ph\u1ea3n \u1ee9ng qPCR, \u0111\u00f3 l\u00e0 ph\u1ed1i tr\u1ed9n c\u00e1c th\u00e0nh ph\u1ea7n \u0111\u01a1n l\u1ebb ho\u1eb7c s\u1eed d\u1ee5ng qPCR mix (c\u00e1c th\u00e0nh ph\u1ea7n \u0111\u01b0\u1ee3c ph\u1ed1i tr\u1ed9n s\u1eb5n th\u00e0nh m\u1ed9t lo\u1ea1i dung d\u1ecbch)<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>3.1. M\u1eabu (khu\u00f4n DNA\/RNA)<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Th\u00f4ng th\u01b0\u1eddng, m\u1eabu tr\u01b0\u1edbc khi \u0111\u01b0a v\u00e0o ph\u1ea3n \u1ee9ng khu\u1ebfch \u0111\u1ea1i c\u1ea7n \u0111\u01b0\u1ee3c x\u1eed l\u00fd m\u1eabu v\u00e0<strong><a href=\"http:\/\/ihhnv-virus-gay-benh-hoai-tu-co-quan-tao-mau-va-co-quan-lap-bieu-mo\"> t\u00e1ch chi\u1ebft DNA\/RNA<\/a><\/strong>.
